Proteomic Antibodies
Anti-peptide antibodies for use in mass spec assay workflows
Antibodies to tryptic (or other enzyme derived) fragments present an excellent solution for highly accurate and quantifiable measurement of protein targets isolated from biological samples. We at Legacy have been at the forefront of method and protocol development for the creation of these reagents for the last 15 years, making thousands of critically useful tools for various applications.
Our Methods
The process for this essentially follows our Core Polyclonal service, yet there are some key differences:
- The client typically provides the target sequence(s) based on internal MS results rather than have us perform antigen design. We are happy to make recommendations pertaining to antigenicity or sequence difficulty to help narrow down the scope as needed.
- Affinity purification includes a number of additional steps to ensure the resulting antibodies are ‘clean’, i.e., with very low background signal.
- A heavy isotope labeled version of the peptide is used for affinity purification, albeit one different from the antigen peptide or heavy standard typically used in the final assay.
- Custom heavy and light peptide standards can be produced and added to the order as needed.
Advanced Needs
Just as with any polyclonal project, it is possible to progress your project to a rabbit monoclonal if you need a more permanent form of the reagent to carry forward into higher scales or more secured production. It is beneficial – but not required – to know this at the onset of the project, as early prep towards this end can avoid potential delays later in the process.
