Monoclonal Antibodies

A step up

Monoclonals offer the potential for a number of improvements over polyclonals should the requirements demand.  Whereas the latter are a collection of all the antibodies made as a response to an antigen, with the former you can continuously screen clones until finding the one that works best for you.  This clone can then be expanded to produce the same antibody as long as it is required, a huge benefit over the potential lot-to-lot variability found in polyclonals. 

Our monoclonal offerings consist of two possible avenues:

Mouse Hybridoma

  • Phase 1: Injection of mice, test bleed with ELISA
  • Phase 2: Splenectomy of selected animal, fusion of splenocytes with myeloma cell line, multi-stage screen to isolate positive clones
  • Phase 3: Subcloning of selected parentals to isolate monoclones with stable reactivity
  • Phase 4 (optional): Expansion and purification at desired scale
  • Phase 5 (optional): Sequencing of selected clones

Rabbit B-cell sorting

  • Phase 1a: Essentially a typical Core Package polyclonal
  • Phase 1b: Additional confirmatory ELISA, options for splenectomy of selected animal or whole blood collection, isolation of splenocytes/PBMCs
  • Phase 2: Single cell sorting to isolate positives
  • Phase 3: Variable region cloning, transient expression as LEM, ELISA
  • Phase 4 (optional): Sequencing of VH/VL chains, stable expression, in mammalian cell line, small scale purification by protein A, ELISA and SDS-PAGE
  • Phase 5 (optional): Scale up to 1L production, purification by protein A, SDS-PAGE, final QC packet

How to decide

Mouse projects average 5-6 months while rabbit projects average 6-7 months

Average cost of rabbit project is roughly 2x that of mouse

Typical mouse project ends with cell line to be stored, while rabbit nets a purified recombinant with known sequences

Rabbits have a more robust immune response than mice, and a higher number of initial clones can be tested
